Fagomine Communication and Immunology: An alkaloid anorectic

Researchers at the Higher Council for Scientific Research Spain have developed a biobased method to mass produce the fagomine, a new compound to fight against the feeling hunger.

The fagomine is an alkaloid, piperidine core, present in small amounts in the seed of buckwheat. By delaying postprandial absorption of carbohydrates (simple and xomplexes), it also delays the recurrence of hunger, due to the rapid drop in blood sugar.

It will be produced in large quantities with the use of recombinant enzyme. And could be marketed at the end of the year in the U.S..

Tissue Repair and Wnt protein

Team: Stanford (California)
Journal of Translational Medicine

Wnt protein by activating the stem cells of the bone is the origin of the resconstruction bone after a fracture, for example. Researchers have succeeded in improving the speed of fracture healing in mice genetically modifying them to over-activate the Wnt pathway.

Moreover, by administering the protein Wnt3 in liposomes in normal mice (vectorization), they were able to increase the speed of their bone repair without generating excessive formation of bone that was feared. This study opens

clinical pathways in the repair of fractures, but also of stroke, heart attacks or skin lesions.

VGX100: An anticancer antibodies against VEGF

VEGF (Vascular Endothelial Growth Factor) is a growth factor necessary for the development of vessels in a tumor (among others). The VGX100, developed by Circadian Technologies Limited, which specializes in antibodies directed against VEGF, is an inhibitor of this factor.

The results of the study to be presented soon show that in combination with docetaxel (an antimitotic agent used in chemotherapy) and bevacizumab (Avastin, also antibody against VEGF), this drug offers survival 2.7 times higher in mice bearing prostate tumors the study period, compared to the association without VGX100. An interesting statistic is that the combination of three drugs enabled 40% of the mice finished the study without tumor, against 0% in other cases.

The worldwide turnover of Avastin, owned by Roche, is almost U.S. $ 9 billion, which shows the potential of this new Antibodies: VGX100.

Gene Therapy : Protein targeting tumors

Team: University of Leeds (UK)
Review: Gene Therapy

Researchers at Leeds University have shown, in vitro, by integrating a virus protein targeting a marker expressed by tumor cells of the bladder (and possibly other cancers), that selectively penetrated into these tumor cells. They also showed that by integrating these proteins ("retargeting proteins") to an existing viral vector for gene therapy, they increased the sensitivity of tumor cells to chemotherapy cancer.

cancer gene therapy, somewhat maligned in recent years because of its lack of specificity vis-à-vis the cancer cells would thus, in humans, increase the effectiveness of chemotherapy, or even repair genetic modifications to the origin of tumors.

The interest of this study is to increase the selectivity of viral vectors in gene therapy, and is successful enough, apparently. It is also possible, thanks to this discovery, target multiple tumor types with the same vector, simply by integrating multiple protein targeting. Similarly, by analyzing the surface antigens of the patient's tumor, it could "build" a specific virus, expressing the target proteins effective against the tumor.

vaccine against type 1 diabetes

Team: University of Calgary, Canada
Journal: Immunity (U.S.)

Type I diabetes is an autoimmune disease. Cytotoxic T lymphocytes attack the beta cells in the pancreas that produce insulin, recognizing a surface antigen of these cells. (Si quelqu'un connait le nom du peptide, je prends!).

The vaccine developed and tested on mice is made up of nanoparticles that have the peptide in question. This has the effect of stimulating T reg cells, suppressor, whose function is to inhibit the cytotoxic response responsible for diabetes.

trials on humans could begin within two years, but may not be as successful because of the large differences between systems immune human and mouse.
This remains a great news, because in addition to being a real bastard, diabetes (I and II) is the source of 12.5 billion euros (2.5 billion for Sanofi insulin) of annual expenditure for safety, with 5 billion more for indirect costs.

aging population and lack of health insurance

A review of the April 22, 2010 HCAAM (High Council on the future of health insurance) provided an update on the impact of aging in the deficit of health insurance.

The notice confirms some accepted ideas and crippled others. For example, the cost of care is actually an increasing function of age. The population in category age over 60 years, is responsible for 45% of health spending, and that in the category over 75 years, 8% to 20% of health spending. However, the idea that the majority of expenses for care of a person is in the final year of his life is wrong: spending the last year of life represents only 7-8% of total granted during a life.
HCAAM The expected demographic aging will contribute 0.4 points to growth in health spending of health insurance, a figure roughly equivalent to the growth of ALD (long Disorders duration).

In conclusion, HCAAM considers "a clear policy of finding more efficient care pathways which will both reduce the impact of demographic factors on health insurance spending and the wider health and provide supported more consistent with the needs and wishes of senior citizens, which include rupture with the home are traumatic in nature. Cells MUSE: new pluripotent stem cells!

A Japanese team from Tohoku University has discovered a new class of pluripotent stem cells called MUSE: Differentiating Stress Enduring Multilineage Cell.

The different categories of stem cells are totipotent cells
  • : Can make an entire organism + placenta. These are the cells of the first 4 days of the embryo.
  • pluripotent cells : Can manufacture all types tissue, but not the placenta. Eg embryonic stem cells.
  • multipotent cells: Several types of cells. Eg hematopoietic stem cells (HSC).
  • unipotent Cells May not be differentiated in a single cell type. Ex: CFU-E (~).
Pluripotent cells are useful for regenerative medicine because they allow the recovery of any organ, a few technical details. Currently, we knew that in humans two types of pluripotent stem cells: embryonic stem cell (ESC) and adult stem cells reprogrammed (IPS), discovered in 2007 by researchers at Kyoto University. Both types have often been tested in studies of regenerative medicine, but often give birth, in addition to fabrics, to tumors greatly limiting their use.

MUSE cells derived from bone marrow. They are rather rare: 1 in 5000. They coexist with CSH, which are only multipotent, let us remember. They were discovered by "serendipity" in error by introducing an enzyme into a toxic skin cell culture, only cells MUSE survived. The researchers have analyzed and made this important discovery, even fundamental to my eyes!

tests for cell differentiation MUSE have been satisfactory, they gave birth to several tissues: muscle, nerve, liver in mice ... Moreover, implanted into the testes of rats, they gave no tumor after 6 months, while ESC and iPS give in after 8 weeks. This property is probably very promising due to their limited proliferation potential, only 2 weeks in culture, against an indefinite multiplication for the two other pluripotent cell types.

prospects of this discovery in the years ahead are considerable, especially in the field of cell therapy.

Reform U.S. Health System: Drug Price Increase

reform U.S. health care system, promised by Obama, has opened a huge market for large pharmaceutical companies: a new access care for millions of people.
In return, these groups have given discounts on the prices of drugs sold in large boxes of federal support for health care.

But the report of Express Scripts announced an unprecedented increase in drug prices United States 10 years: 9% in one year. The biotech drugs increased 11.5%. These increases reflect a desire to pass on the cost reductions mentioned previously.

79-6, promising new compound in lymphomas

Team: International led by the Ontario Cancer Institute
Journal: Cancer Cell (13 / 04 U.S.)

BCL-6 protein was identified as the source of over half of non-Hodgkin lymphoma, and many forms of diffuse large B cell lymphoma

We know that blocking this protein, BCL-6 by knockout experience for example, kills tumor cells of diffuse large B cell lymphomas This protein was therefore an attractive therapeutic target. However, as a transcription factor, it acts through protein-protein interaction, lockable hard by a small molecule.

However, after its three-dimensional structure elucidated by crystallography 3D, the researchers carried out a docking of the molecule with more than a million compound (virtual simulation of the interaction between the molecule and the compound) and were able to discover 79-6.

Tested on mice bearing human lymphoma cells, 79-6 decreased very significantly the tumor cells with low toxicity. It is therefore an attractive therapeutic compound.

CCL21 protects tumor immune

Team: Polytechnic LausanneRevue: Science (U.S., March 2010)

The researchers studied tumor cells melanoma in mice. They identified that they secrete a protein: Chemokine Ligand 21 (CCL21, also known 6Ckine). This chemokine is expressed in lymph node-level to protect an autoimmune response by leukocytes.
CCL21 thus allows the tumor to evade the immune response, by "mimicking" a lymph node. This discovery could have impacts on long-term immunotherapy.

Telcagepant: A new drug against migraine

Journal: Lancet
Team: Unspecified. "Lars Edvinsson (Lund University Hospital, Sweden) and Mattias Linde (St Olavs Hospital, Trondheim, Norway) "?

Telcagepant (formerly MK-0974, developed by Merck (U.S.)) acts on CGRP (Calcitonin gene-related peptide) . CGRP is a neuropeptide such as substance P, endorphins, neuropeptide Y (NPY). Neuropeptides generally act on GPCRs coupled receptors (G-protein) and have a medium-term modulatory action on neurons.

studies suggesting that a migraine attack is associated with release of CGRP from meningeal nociceptors in the trigeminal ganglion, the idea an antagonist of CGRP receptors directly came to the researcher, and Telcagepant is one. The effectiveness of this compound appears to be similar to triptans (which are currently used treatment of crisis, and act at serotonin receptors). Gold triptans, which in addition to being relatively expensive (Zomig: 30 € 6CP box), are formally cons-indicated in case a history of heart attack or angina pectoris, of history of stroke or peripheral, certain cardiac arrhythmias, or uncontrolled hypertension, as strongly vasoconstrictors, not to mention interactions with dihydroergotamine. And Telcagepant escapes these cons-indications and interactions, because there is no vasoconstrictor.

In contrast, phase II trials were halted in March 2009 because of liver toxicity apparently too strong, with high elevation of transaminases. It is on this toxicity is that the assessments currently being conducted by Merck. Moreover, the telcagepant induced side effects: nausea, dizziness, numbness or tingling skin.

Migraine affects 15% of the adult population in France.

Zolmitriptan (antimigraine major) is a global turnover of $ 158M in 2008.

CRP increased in colon cancer

Team: Vanderbilt University, Tennessee, U.S.
Review: American Association for Cancer Research

The researchers studied 338 cases of cancer and 451 healthy women, and measured blood levels of CRP, protein used as a marker of inflammation. The result is that the quartile of highest values had a 2.5 times higher risk of having cancer colon than the lowest quartile.

A marker exists for colorectal cancer, but it is not used in the diagnosis, because its specificity is not perfect, it is the ACE protein. The specificity of CRP is obviously not better because it is increased in many circumstances.

New patent for a sublingual dosage form

  3. Better absorption (bioavailability superior in most cases, the oral even for non-hepatic metabolism)
  4. Avoidance of possible gastrointestinal toxicity
Note that the points to reduce the dose of the medication have included an environmental impact as well, decreasing the doses logically rejected in nature, and therefore the pharmaceutical pollution.

The problem that researchers are still working for an SME dijonaise associated Bionetwork, is the masking of bad taste sometimes annoying active ingredients.

Illustration: Different curves oral bioavailabilities as Tmax. In the sublingual, the curve can be likened to that of a mouth with a Tmax very low (approaching the dark blue curve)

Genetic Diagnosis: Patentability of genes

A U.S. federal judge recently struck down patents both BRCA1 and BRCA2 (Breast Cancer 1 and 2). These two genes are tumor suppressor genes, some mutations have been identified as the cause of breast and ovarian cancer.

This decision calls into question the exclusive rights to market the Myriad BRACAnalysis test: This test can detect cancer, hereditary breast and ovarian cancer hereditary with a confidence rating of 82% in the first case and 44 % in the second, at a cost of about $ 2000.

The reason given is that the mutated gene, so that-which is based on the test, is not fundamentally different from natural gene, and therefore can not be patented.

However, although 20% of the human genome is patented at the moment, this ruling should be inconsequential, because probably overturned on appeal.

: effect cardiovascular therapeutics?

Study: Modena (Italy)
Journal: Circulation (Cardiovasulaire) (U.S.)

The melanocortin (or rather, because there are several forms) is a peptide hormone, derived from the cleavage central (hypothalamus) of proopiomelanocortin (POMC), under the action of leptin. His action, linked to 5 different receivers is twofold: at the central level, it inhibits food intake and peripheral level, it stimulates melanin synthesis by cutaneous melanocytes.

Italian researchers have just proven its significance in ischemic diseases such as myocardial infarction and stroke apoplectic. The altered production of this hormone is indeed a cause inflammatory aspects of cerebral ischemia or infarction associated with these diseases.
Thus, an intervention, even hours after the accident could take place and allow functional recovery, at least in part, the affected organ.

Much research is underway in the world on the role and employment potential of melanocortin in therapy. No molecule is currently using this pharmacological means.
